The Biological Resources Branch within the National Institute on Aging's Division of Aging Biology maintains colonies of barrier-raised, Specific Pathogen-Free (SPF) male and female mice and rats under contractual arrangement with Charles River Laboratories. These animals are for research that is explicitly on aging and are not a source of adult animals for other areas of research. Limited numbers of animals from the NIA aged rodent colonies are available to investigators with active grants at academic and non-profit research institutions under the terms described on the Eligibility Criteria page. They are available free of charge (including shipping) and must be ordered through the Rodent Ordering System (see Ordering Instructions, Shipment of Animals, and Replacement Policy for more information).
NIA and CRL are taking all necessary steps to identify the source of this contamination and ensure that this unprecedented event does not happen again. The precise duration of the contamination in R17 is not known, but because health monitoring for each barrier room housing the NIA aged rodent colonies is conducted on a monthly basis by CRL and a third-party health monitoring service, we estimate the window for entry of P. carinii into the barrier room after June 15, 2022. Investigators that received rats from R17 after June 15, 2022 have been contacted directly.
R17 is the only barrier affected. Two additional production colonies (K81 in Kingston, NY and R05 in Raleigh, NC) are still available to provide aged rats and it is not anticipated that elimination of R17 will interrupt availability.
Heterochronic blood exchange (HBE) has demonstrated that circulating factors restore youthful features to aged tissues. However, the systemic mediators of those rejuvenating effects remain poorly defined. We show here that the beneficial effect of young blood on aged muscle regeneration was diminished when serum was depleted of extracellular vesicles (EVs). Whereas EVs from young animals rejuvenate aged cell bioenergetics and skeletal muscle regeneration, aging shifts EV subpopulation heterogeneity and compromises downstream benefits on recipient cells. Machine learning classifiers revealed that aging shifts the nucleic acid, but not protein, fingerprint of circulating EVs. Alterations in sub-population heterogeneity were accompanied by declines in transcript levels of the pro-longevity protein, α-Klotho, and injection of EVs improved muscle regeneration in a Klotho mRNA-dependent manner. These studies demonstrate that EVs play a key role in the rejuvenating effects of HBE and that Klotho transcripts within EVs phenocopy the effects of young serum on aged skeletal muscle.

To understand the role of PLA2G2D expression and PGD2/PTGDR signalling in the clinical setting, we first analysed the literature for studies of age-dependent changes in lung levels of PGD2 synthases (PGD2 synthase (PTGDS) and haematopoietic prostaglandin D synthase), the enzymes required for conversion of prostaglandin H2 to PGD2, Ptgdr and Pla2g2d mRNA transcripts. Using RNA-sequencing data from 103 healthy lungs in a study of fibrosis40 (Gene Expression Omnibus (GEO) accession GSE150910) and 578 lungs from the Genotype-Tissue Expression (GTEx) portal41, we found that levels of PTGDS and PTGDR increased with ageing (Fig. 4a, b). Next, we obtained peripheral blood mononuclear cells from young and aged human individuals and cultured them as described in the Methods to obtain dendritic cells. After 14 days, we infected these cells with SARS-CoV-2. SARS-CoV-2 causes an abortive infection of human dendritic cells42. We then measured mRNA expression levels of PLA2G2D, PTGDR and PTGDS and cell supernatant levels of PGD2-methoxime (PDG2-MOX), a stabilized version of PGD2, and 11β-prostaglandin F2α (11β-PGF2α), a PGD2 enzymatic product. Expression of PLA2G2D and PTGDR increased with ageing and that of PLA2G2D further increased after SARS-CoV-2 infection (Fig. 4c). PGD2 increased with ageing and SARS-CoV-2 infection further enhanced expression by dendritic cells from aged individuals (Fig. 4d). 11β-PGF2α increased in dendritic cells from aged people in response to infection (Fig. 4d).

Design, Setting, and Participants This prospective cohort study was part of the Coronary Artery Risk Development in Young Adults (CARDIA) study. Participants were aged 38 to 50 years and wore an accelerometer from 2005 to 2006. Participants were followed for a mean (SD) of 10.8 (0.9) years. Data were analyzed in 2020 and 2021.

Marijuana use continued to rise among college students over the past five years and remained at historically high levels among same-aged peers who are not in college in 2020, according to survey results from the 2020 Monitoring the Future (MTF) panel study. This represents the highest levels of marijuana use recorded since the 1980s. The survey also found that marijuana vaping and nicotine vaping leveled off in 2020 after sharp increases reported every year since 2017 for both college students and same-aged respondents who are not in college.
In addition, daily or near daily use of marijuana has continued to rise among college students since 2015, with 8% of college students reporting using marijuana on a daily or near daily basis in 2020 compared to nearly 5% in 2015. In comparison, 13% of same-age adults not in college reported using marijuana on a daily or near daily basis in 2020, consistent with recent years.Vaping marijuana and nicotine: The 2020 MTF survey also recorded a halt in the dramatic increases in vaping marijuana and vaping nicotine among college-aged adults observed between 2017 and 2019. During this time, the percentage of college-aged adults who reported vaping marijuana in the past 30 days more than doubled, from 5% to 14% for college students, and 8% to 17% for noncollege respondents. In 2020, these increases leveled off, with 12% of college students and 14% of noncollege respondents reporting vaping marijuana in the past 30 days. Similarly, there was a nonsignificant change for vaping nicotine in 2020, with 19% of college students and 24% of noncollege respondents reporting vaping nicotine in the past 30 days compared to 22% and 18% in 2019, and 6% and 8% in 2017, respectively.
